02.11 (2)

Spent the most of the day writing the intro, and a minor, half assed part of the day writing the previous day's summary of what was done before november

This one will be focused on a small (more detailed) recap of what I have managed to achieve since restarting my labwork around the second week of november.

RNA extraction from collected plant tissue

measuring the concentrations on nanodrop

DNAsing (DNA digest)

validation of DNAsing

cDNA synthesis

Last friday I repeated the DNA digest, and now I think I will get back onto track with seeding the new in vitro population.

I botched the cDNA synthesis because I didn't know how to pipette things reliably (I hopefully know now I will write my notes on it here) and also my micropipette started breaking down and was unresponsive and you need specific equal concentrations of RNA for cDNA syntehsis.

All in all despite some my technical replicates failing which causes me and my supervisor undue stress due to my middling pippetting skills and issues with focusing for a longer perioids of time, and that preventing me from moving forwards I am happier about this stage of the labwork, I feel more confident and I when I am left to do something alone (run a PCR, gel imaging, prepare a mix) it pans out more often than before, and I know if something works out in days instead of weeks. I am getting better but the time is tight

same as 01.11 Will add some photos, tables and details tmrw because I am so tired, my plans for tommorow are reading and understanding two papers about current understanding of mlo11 that I will probably let some web enabled llm on lmarena pick them for me (I don't like using what chatbots write it feels nasty but if they give me a link to a human written article I don't mind)
